Please also check whether the Integrated Windows authentication was checked. If not, please check it. Then in the Permission Groups, make sure the Exchange servers is checked.
If the issue persists, please first ensure we can resolve the FQDN of the Hub server by using ping command.
The most likely cause of this issue is the third party security software which blocks the communication between the server.
Thus, please use telnet to send the email to Hub server from Edge server on port 25. Check whether the email could be delivered.
And please paste the information that you get when running the telnet command in order to find the root cause of the issue.
